One of the most promising avenues of research in immune oncology is long-term, live imaging. Capturing dynamic biological processes in real time holds the key to understanding which cell-cell interactions promote a successful immune response, and which promote disease.
Feature application: Cell Metabolism
In this Feature Application, we show that: (1) holotomography can resolve the spatial and temporal dynamics of mitochondria, LDs and mitochondria-LD interactions in unperturbed conditions; (2) it is possible to induce mitochondrial dysfunction and then restore functioning by the addition of a specific drug and (3) it is possible to induce and then visualize the early stages of LD formation.
Feature application: Characterisation of Stem Cells
In this Feature Application, we show how Nanolive’s technology can be used to: (1) observe MSCs in 3D over long periods of time at high spatial and temporal resolution; (2) capture mitotic division events in MSCs in unparalleled detail; (3) observe the dynamics of stress fiber formation and (4) visualize the morphological changes that occur during stem cell differentiation.

In the first case study, we analyze micro-heterogeneity at the population and the temporal level in unperturbed cells and in the second and third case studies we focus on cells where intracellular trafficking and respiration were experimentally perturbed.
Feature application: Characterisation of Neurons
This Feature Application shows the huge potential that Nanolive cell imaging holds for neurobiological research. Our first case study shows a timeline of the morphological changes undifferentiated primary cortical neurons undergo after exposure to neurite stimulation media. High precision segmentations are used to calculate cell metrics (e.g. volume, shape and dry mass) of interest. These calculations are directly linked to novel behaviours observed in individual neurons.
